为了探讨姜黄素对白血病细胞的杀灭能力, 运用细胞培养、流式细胞仪和末端TdT酶标记技术系统研究其药理作用和方式。结果发现, 姜黄素能选择性抑制急性髓性白血病细胞HL-60 的增殖, 呈时间与剂量依赖曲线, 姜黄素体外抑制HL-60 细胞24 h 达到最大峰值。亚G1 凋亡峰出现在12 h 后,并随时间延长而逐步增加,24 h 可达34.4% 。用末端TdT酶标记法进一步证实凋亡的细胞数在同一时相可达到41% 。结果提示: 姜黄素有明确的抗白血病细胞增殖的能力, 其作用机理是诱导细胞凋亡, 体外浓度和作用时间可作为其进一步临床运用的参考。
To investigate the killing ability of curcumin on leukemia cells, cell culture, flow cytometry and terminal TdT enzyme labeling techniques were used to systematically study its pharmacological actions and methods. The results showed that curcumin could selectively inhibit the proliferation of HL-60 cells in acute myeloid leukemia in a time- and dose-dependent manner. Curcumin inhibited HL-60 cells in vitro to reach their maximum peak at 24 h. The sub-G1 apoptotic peak appeared after 12 h, and gradually increased with time, reaching 34.4% at 24 h. Using terminal TdT enzyme labeling, it was further confirmed that the number of apoptotic cells reached 41% at the same time. The results suggest that curcumin has a clear ability to proliferate anti-leukemic cells, its mechanism of action is to induce apoptosis, in vitro concentration and effect time can be used as reference for further clinical application.
