目的探讨丹溪痛风方(DGP)对胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白的影响。方法胰腺癌BxPC-3细胞随机分成模型组,5-氟尿嘧啶(5-FU)组(50μg/ml),DGP高、中、低剂量组(40,20,10μg/ml)。采用台盼蓝染色、MTT法、细胞划痕法分别检测胰腺癌BxPC-3细胞生长曲线、增殖抑制率、运动能力;采用免疫组织化学、RT-PCR法检测胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1水平。结果与模型组比较,5-FU组,DGP高、中剂量组胰腺癌BxPC-3细胞OD值明显降低(P<0.05);运动能力明显降低(P<0.05);Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1水平明显降低(P<0.05);Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1 mRNA水平明显降低(P<0.05)。结论 DGP对胰腺癌BxPC-3细胞Hedgehog信号通路关键蛋白Shh、Smo、Ptch1、Gli1具有抑制作用,可见DGP在胰腺癌治疗中扮演重要角色。
Objective To investigate the effects of Danxi Gout (DGP) on the key proteins of Hedgehog signaling pathway in pancreatic cancer BxPC-3 cells. Methods Pancreatic cancer BxPC-3 cells were randomly divided into model group, 5-fluorouracil group (50μg / ml), DGP high, medium and low dose groups (40,20,10μg / ml). The pancreatic cancer BxPC-3 cell growth curve, proliferation inhibition rate and exercise capacity were detected by trypan blue staining, MTT assay and cell scratch assay respectively. Hedgehog signaling pathway of pancreatic cancer BxPC-3 cells was detected by immunohistochemistry and RT-PCR Key proteins Shh, Smo, Ptch1, Gli1 levels. Results Compared with the model group, the OD value of BxPC-3 cells in 5-FU group and DGP high and medium dose groups was significantly decreased (P <0.05), and the exercise capacity was significantly decreased (P <0.05). The key proteins of Hedgehog signaling pathway Shh, The levels of Smo, Ptch1 and Gli1 were significantly decreased (P <0.05). The levels of Shh, Smo, Ptch1 and Gli1 were significantly decreased in Hedgehog signaling pathway (P <0.05). Conclusion DGP inhibits the key proteins Shh, Smo, Ptch1 and Gli1 of Hedgehog signaling pathway in pancreatic cancer BxPC-3 cells, indicating that DGP plays an important role in the treatment of pancreatic cancer.
