目的 :研究从胆囊粘膜典型增生到胆囊癌的发展过程中肿瘤坏死因子 (TNF)的表达 ,进一步探讨它们在胆囊癌发生、发展中的意义。方法 :用原位杂交及免疫组织化学方法检测胆囊结石引起的粘膜典型增生、非典型增生及胆囊癌的TNFmR NA、TNF蛋白的表达。结果 :①TNFmRNA在胆囊粘膜典型增生、非典型增生及胆囊癌组织上皮细胞或癌细胞和单个核细胞(MNC)的阳性表达率及表达细胞数目均逐级增高。②在胆囊癌组织中 ,癌细胞及MNC表达TNFmRNA的细胞表达数目随肿瘤进展而增多 (P <0 .0 1 )。③TNF蛋白与TNFmRNA表达区域相似 ,阳性表达率较高。④癌细胞和MNC的TNFmRNA及蛋白表达均呈正直线相关。结论 :TNF参与了胆囊结石致癌的发病过程 ,并与胆囊癌的进展有关。
Objective : To study the expression of tumor necrosis factor (TNF) during the development of gallbladder carcinoma from typical hyperplasia to gallbladder carcinoma, and further explore their significance in the occurrence and development of gallbladder carcinoma. METHODS: TNFmR NA and TNF protein expressions in mucosal hyperplasia, atypical hyperplasia, and gallbladder carcinoma were detected by in situ hybridization and immunohistochemistry. RESULTS: 1The positive rate of TNF mRNA expression in epithelial cells or cancer cells and mononuclear cells (MNC) of gallbladder mucosal proliferation, atypical hyperplasia, and gallbladder cancer was gradually increased. 2 In gallbladder carcinoma tissues, the number of TNF mRNA-expressing cells in cancer cells and MNCs increased with tumor progression (P < 0.01). 3 The TNF protein and TNF mRNA expression regions are similar, and the positive expression rate is higher. 4 The expression of TNF mRNA and protein in cancer cells and MNC were positively correlated. Conclusion : TNF is involved in the pathogenesis of carcinogenesis of gallbladder stones and is associated with the progression of gallbladder cancer.
