目的:观察转化生长因子β1(TGF-β1)作用下绒癌JEG-3细胞的增殖及其Smad3,7 mRNA的表达变化,进一步探讨TGF-β/Smads信号通路在绒癌发生发展中的作用机制。方法:分别取不同浓度和不同作用时间的TGF-β1处理绒癌JEG-3细胞,采用四甲基氮唑法(MTT)检测细胞的增殖活力,RT-PCR技术检测Smad3,7 mRNA的表达差异。结果:TGF-β1促进了绒癌JEG-3细胞的增殖;随着TGF-β1应用浓度的增高,Smad3 mRNA的表达水平逐渐升高,用一定浓度的TGF-β1作用12h后Smad3 mRNA的表达量随着时间的延长开始逐渐增加;同样条件下Smad7 mRNA的表达差异无统计学意义。结论:TGF-β1可促进绒癌JEG-3细胞的增殖;Smad3mRNA对外源性的TGF-β1有良好的反应性,Smad7 mRNA则表现为无应答。
OBJECTIVE: To observe the proliferation and the expression of Smad3,7 mRNA in choriocarcinoma JEG-3 cells induced by transforming growth factor-β1 (TGF-β1), and to explore the mechanism of TGF-β / Smads signaling pathway in carcinogenesis and progression of choriocarcinoma . Methods: The choriocarcinoma JEG-3 cells were treated with TGF-β1 at different concentrations and different time, respectively. The proliferation of JEG-3 cells was detected by MTT assay. The expression of Smad3,7 mRNA was detected by RT-PCR . Results: TGF-β1 promoted the proliferation of choriocarcinoma JEG-3 cells. With the increase of TGF-β1 concentration, the expression of Smad3 mRNA gradually increased. After a certain concentration of TGF-β1 was treated for 12 hours, the expression of Smad3 mRNA With the extension of time began to gradually increase; under the same conditions Smad7 mRNA expression was no significant difference. CONCLUSION: TGF-β1 can promote the proliferation of choriocarcinoma JEG-3 cells. Smad3 mRNA has good reactivity to exogenous TGF-β1 and Smad7 mRNA shows no response.
