目的:研究对羟基苯甲酸丁酯对雄性小鼠性成熟前期的睾丸毒性。方法:对出生后3天小鼠皮下注射对羟基苯甲酸丁酯,建立中毒小鼠模型,染毒剂量如下:随机分成五组:空白生理盐水对照组A组,阳性雌激素对照组B组,实验组C组[对羟基苯甲酸丁酯钠低、中、高剂量组(L100mg/kg、M200mg/kg、H400mg/kg组)]颈部背侧皮下染毒,连续染毒60天后处死小鼠,通过精子分析仪观察睾丸每日精子生成量、精子活动度、附睾尾精子计数、精子畸形率;用ELISA法测小鼠血清中睾酮、睾丸组织中SOD的含量;结果:与对照组相比L、M、H组及B组的T和SOD明显降低(P<0.05)。L、M、H组及B组的每日精子生成量、精子活动率、附睾精子计数明显降低,精子畸形率明显升高(P<0.05)。L、M、H组及B组的的睾酮含量均降低(P<0.05)结论:在雄性小鼠性成熟期前长时间给予对羟基苯甲酸丁酯钠干扰,能降低血清T激素水平,同时降低其睾丸组织中SOD活性,导致仔鼠精子数量和精子活动度降低,畸形率增加。
OBJECTIVE: To study the testicular toxicity of butylparaben in premature sexual maturation in male mice. Methods: Three days after birth, mice were injected subcutaneously with butylparaben to establish the model of poisoning mice. The doses of the mice were randomly divided into five groups: blank control group A, positive estrogen control group B, In the experimental group C, mice in the low, middle and high dose sodium butylparaben group (L100mg / kg, M200mg / kg, H400mg / kg) were drenched subcutaneously in the dorsal neck of the neck, The daily sperm production, sperm motility, epididymal sperm count and sperm deformity were observed by sperm analyzer. The contents of SOD in testis and testis were detected by ELISA. Results: Compared with the control group The levels of T and SOD in L, M, H and B groups decreased significantly (P <0.05). The daily sperm production, sperm motility and epididymal sperm counts of L, M, H group and B group were significantly decreased, and the sperm deformity rate was significantly increased (P <0.05). L, M, H and B decreased (P <0.05) .Conclusion: The prolonged administration of butylparaben in male mice before sexual maturation can decrease the level of serum T-hormones, meanwhile, Reduce the activity of SOD in the testis tissue, resulting in the reduction of the number of spermatozoa and sperm motility in offspring and the increase of deformity rate.
