目的进一步探索聚合酶链反应(PCR)试验用于诊断布氏菌病的可行性。方法采用由编码牛种布氏菌31KD同一蛋白基因序列设计的3对引物,扩增片断分别为223bp、330bp和230bp的PCR试验,对急、慢性期布氏菌病患者(50例)、健康人(45例)及感染布氏菌后豚鼠的全血进行检测。结果42例急慢性期布氏菌病患者和5例健康人PCR试验为阳性,感染豚鼠全部阳性。结论PCR试验诊断布氏菌病病人具有较高的敏感性和一定的特异性,其应用前景较好。
Objective To further explore the feasibility of polymerase chain reaction (PCR) assay for diagnosis of brucellosis. Methods Three pairs of primers designed from the same protein sequence of 31KD of Bovine Escherichia coli were used to amplify the PCR products of 223bp, 330bp and 230bp, respectively. The results showed that there were 50 patients with acute and chronic brucellosis, (45 cases) and whole blood of guinea pigs infected with brucella. Results 42 cases of acute and chronic brucellosis patients and 5 healthy people PCR test was positive, all infected guinea pigs were positive. Conclusion PCR test diagnosis of brucellosis patients with high sensitivity and a certain specificity, its application prospects are better.
