目的:研究纤体降脂Ⅰ号对去卵巢大鼠下丘脑瘦素信号传导通路的影响,探讨其纤体降脂的作用机制。方法:72只200 g左右的SD大鼠按体质量随机抽出假手术组12只,造模组60只,造模组采用双侧去卵巢法造模,假手术组只切除卵巢周围相应质量的脂肪。造模成功后,造模组大鼠按体质量随机分为模型组、纤体降脂Ⅰ号高剂量组(13.2 g/kg)、中剂量组(6.6 g/kg)、低剂量组(3.3 g/kg)和戊酸雌二醇组,每组12只。术后第5天开始药物干预,模型组和假手术组给予相应体积的纯净水,持续13周。给药结束后,取下丘脑匀浆,酶联免疫法检测瘦素(Leptin,LP)、瘦素长形受体(OB-Rb)、神经肽Y(NPY)、α-促黑激素(α-MSH)含量。结果:纤体降脂Ⅰ号能够显著增加OB-Rb及αMSH的含量,降低NPY的含量。结论:通过增加下丘脑OB-Rb的表达,抑制NPY的合成与分泌,促进αMSH的合成,减少摄食,增加能量消耗,抑制脂肪形成,可能是纤体降脂Ⅰ号纤体降脂的作用机制之一。
OBJECTIVE: To study the effect of slipping fat No.1 on the hypothalamic leptin signal transduction pathway in ovariectomized rats and to explore its mechanism of action of slimming lipid. Methods: Seventy-two SD rats weighing about 200 g were randomly divided into sham operation group (n = 12) and model group (n = 60). The model group was made by bilateral ovariectomy. fat. After successful modeling, the rats in model group were randomly divided into model group, high-dose group (13.2 g / kg), medium dose group (6.6 g / kg) and low dose group g / kg) and estradiol valerate group, 12 in each group. The first 5 days after the start of drug intervention, the model group and sham group were given the appropriate volume of pure water for 13 weeks. After administration, the hypothalamus was removed and the levels of leptin (LP), OB-Rb, NPY, α -MSH) content. Results: Slimming lipid-lowering Ⅰ can significantly increase the content of OB-Rb and αMSH, reduce the content of NPY. CONCLUSION: The mechanism of hypolipidemic effect of Slimming Hypothyroidism Ⅰ may be through increasing the expression of OB-Rb in the hypothalamus, inhibiting the synthesis and secretion of NPY, promoting the synthesis of αMSH, decreasing food intake, increasing energy consumption and inhibiting adipogenesis one.
