目的:检测骨化三醇对人宫颈癌SiHa细胞维生素D受体(VDR)及S期激酶相关蛋白2(Skp2)表达的影响,探讨其抗癌机制。方法:选用人宫颈癌SiHa细胞进行体外培养。CCK-8法测定骨化三醇对细胞生长抑制率的影响;流式细胞仪进行细胞周期分析;Western blot法检测VDR及Skp2表达的变化。结果:(5×10-8~10-6)mol/L骨化三醇作用于SiHa细胞1~4天,细胞生长增殖均受到显著抑制;且随着作用浓度增加,细胞增殖能力逐渐下降。10-7mol/L骨化三醇作用于SiHa细胞后,G0/G1期细胞比例增加,S、G2/M期细胞比例降低,细胞增殖指数(PI)降低(P<0.05);VDR蛋白表达增加(P<0.05),Skp2蛋白表达显著减少(P<0.01)。结论:骨化三醇对人宫颈癌SiHa细胞的增殖具有显著的抑制作用,其机制可能是通过上调VDR的表达,参与Skp2基因转录的调控,降低Skp2表达。
OBJECTIVE: To investigate the effect of calcitriol on the expression of vitamin D receptor (VDR) and S-kinase 2 (Skp2) in human cervical cancer SiHa cells and to explore its anticancer mechanism. Methods: Human cervical cancer SiHa cells were selected for in vitro culture. CCK-8 method was used to determine the effect of calcitriol on cell growth inhibition; flow cytometry was used for cell cycle analysis; Western blot was used to detect the expression of VDR and Skp2. RESULTS: (1 × 10-8 ~ 10-6) mol / L calcitriol acted on SiHa cells for 1 ~ 4 days, and the proliferation and proliferation of SiHa cells were significantly inhibited. With the increasing concentration of calcitriol, the cell proliferation ability decreased gradually. After treated with 10-7mol / L calcitriol, the proportion of cells in G0 / G1 phase increased, the proportion of cells in S, G2 / M phase decreased, the cell proliferation index (PI) decreased (P <0.05), Skp2 protein expression was significantly reduced (P <0.01). CONCLUSION: Calcitriol has a significant inhibitory effect on the proliferation of SiHa cells in human cervical cancer. The possible mechanism is that it may be involved in the regulation of Skp2 gene transcription and the decrease of Skp2 expression by up-regulating VDR expression.
