目的:建立高效液相色谱法测定风湿定胶囊中欧前胡素和异欧前胡素的含量。方法:Waters e2695 HPLC色谱系统(Empower Waters工作站,Waters 2489 UV/visible Detector检测器,Hypersil ODS2色谱柱(4.6 mm×150 mm,5μm),柱温25℃,流速1.0 mL·min-1,流动相甲醇-水(62∶38),检测波长254 nm,进样量20μL。结果:欧前胡素在1.064~31.92 mg·L-1呈良好的线性关系(r=0.999 7),异欧前胡素在1.090~32.70 mg·L-1呈良好的线性关系(r=0.999 3)。欧前胡素的平均加样回收率100.2%(n=9),RSD 1.47%,异欧前胡素的平均加样回收率为100.0%(n=9),RSD 1.64%,结论:该方法操作简便、准确、稳定、灵敏度高,适用于风湿定胶囊中欧前胡素和异欧前胡素的质量控制。
Objective: To establish a method for the determination of imperatorin and isoimperatorin in Fengshuiding Capsules by high performance liquid chromatography. METHODS: A Waters e2695 HPLC system (Empower Waters workstation, Waters 2489 UV / visible Detector detector, Hypersil ODS2 column (4.6 mm × 150 mm, 5 μm) was used with a column temperature of 25 ℃ and a flow rate of 1.0 mL · min -1. Methanol-water (62:38), the detection wavelength was 254 nm, and the injection volume was 20 μL.Results: Imperatorin had a good linear relationship (r = 0.999 7) between 1.064 and 31.92 mg · L-1, The calibration curve was linear over the range of 1.090 ~ 32.70 mg · L-1 (r = 0.999 3) .The average recovery of imperatorin was 100.2% (n = 9), RSD was 1.47% The average recovery was 100.0% (n = 9), RSD 1.64%. Conclusion: The method is simple, accurate, stable and sensitive. It is suitable for the quality control of imperatorin and isoimperatorin .
