目的观察大建中汤对脾阳虚大鼠大脑中环氧合酶-2(COX-2)mRNA的影响,探讨大建中汤对脾阳虚的作用机理。方法将SD大鼠40只随机分为正常组、模型组、大建中汤大、小剂量组各10只,采用综合造模法制备脾阳虚动物模型,根据实验分组以灌胃形式进行给药,各组大鼠连续给药15天后,断颈处死大鼠并立即取脑组织。用RT-PCR法分析检测脑组织COX-2mRNA表达的变化。结果与正常对照组相比,脾阳虚模型组大鼠大脑COX-2mRNA表达明显增加,具有显著统计学意义(P<0.01);大建中汤高剂量组COX-2表达与模型组相比明显下降,具有显著性差异(P<0.01)。结论 COX-2mRNA在脾阳虚中高表达,大建中汤可抑制脾阳虚大鼠大脑中COX-2的表达,并可能通过此抑制作用来达到治疗脾阳虚证的目的。
Objective To observe the effects of Dajianzhong Decoction on cyclooxygenase-2 (COX-2) mRNA in the brain of rats with spleen yang deficiency and to explore the mechanism of Dajianzhong Decoction on spleen-yang deficiency. Methods Forty Sprague-Dawley rats were randomly divided into normal group, model group and Daijianzhong decoction group, 10 rats in each group. The animal model of spleen-yang deficiency syndrome was established by comprehensive modeling. After 15 consecutive days of administration of each group of rats, the rats were sacrificed and the brains were removed immediately. The expression of COX-2 mRNA in brain tissue was detected by RT-PCR. Results Compared with the normal control group, the expression of COX-2 mRNA in the brain of spleen-yang deficiency model group was significantly increased (P <0.01), and COX-2 expression in high-dose Jianjianzhong group was significantly higher than that in the model group Significantly decreased, with significant difference (P <0.01). Conclusions COX-2 mRNA is highly expressed in spleen Yang deficiency. Dajianzhong decoction can inhibit the expression of COX-2 in the brain of rats with spleen yang deficiency and may achieve the goal of treating spleen-Yang deficiency syndrome.
