目的:观察表达变异链球菌表面蛋白PAcP和霍乱毒素B亚单位融合蛋白(PAcP/CTB)的转基因番茄的免疫原性和安全性。方法:取24 d龄BABL/c小鼠18只,随机分成3组(n=6),分别用转基因番茄果汁(实验组)、变异链球菌灭活全菌(阳性对照组)、非转基因番茄果汁(阴性对照组)灌胃免疫,共4次,每次间隔1周;于首次免疫前1 d和每次免疫后1周称体质量,采集血清、唾液样品,用ELISA法检测血清中IgG、唾液中SIgA抗体水平;最后1次采样后处死所有动物,取心、肝、脾、肺、肾进行组织病理检查。结果:免疫后,实验组和阳性对照组小鼠血清特异性IgG和唾液特异性SIgA水平均明显升高,与阴性对照组相比P<0.05;各组小鼠免疫前后的体质量变化均无统计学差异(P﹥0.05);组织病理学检查,实验组心、肝、脾、肺、肾均未见明显异常。结论:转基因番茄表达的外源目的蛋白PAcP/CTB具有免疫原性和一般安全性。
Objective: To observe the immunogenicity and safety of transgenic tomato expressing PAcP and cholera toxin B subunit fusion protein (PAcP / CTB) of Streptococcus mutans. Methods: Eighteen BABL / c mice of 24 days old were randomly divided into three groups (n = 6), which were treated with transgenic tomato juice (experimental group), streptococcus mutans inactivated whole bacteria (positive control group), non-transgenic tomato Fruit juice (negative control group) were given intragastrically for 4 times at intervals of 1 week. The body weight was measured on the 1st day before the first immunization and 1 week after each immunization. Serum and saliva samples were collected and the serum IgG , SIgA antibody levels in saliva; after the last sampling all animals were sacrificed, and heart, liver, spleen, lung and kidney were taken for histopathological examination. Results: After immunization, the levels of serum specific IgG and salivary specific SIgA in experimental group and positive control group were significantly higher than those in negative control group (P <0.05). The body weight of mice in each group were not changed before and after immunization Statistical differences (P> 0.05); histopathological examination, the experimental group heart, liver, spleen, lung, kidney were no significant abnormalities. Conclusion: The exogenous protein PAcP / CTB expressed in transgenic tomato has immunogenicity and general safety.
