目的探讨4’-甲醚-黄芩素(4-MS)诱导人绒毛膜癌JAR细胞凋亡的分子机制。方法应用透射电镜、流式细胞术、实时定量PCR法和Western blotting方法,体外观察4-MS对JAR细胞的影响。结果电镜下可见用药后JAR细胞凋亡的形态学改变;流式细胞术显示,随着作用时间和药物剂量的增加,早期凋亡细胞和坏死细胞增加,明显高于对照组(P<0.01)。20、40 mg.L-1 4-MS作用48 h后c-myc mRNA的表达量为0.87±0.12和0.45±0.08;c-myc蛋白的表达量是0.37±0.02和0.25±0.02,均低于对照组(P<0.01)。结论 4-MS诱导JAR细胞凋亡,其机制与降低c-myc的表达有关。
Objective To investigate the molecular mechanism of apoptosis induced by 4’-methyl-baicalein (4-MS) on human choriocarcinoma JAR cells. Methods The effects of 4-MS on JAR cells were observed by transmission electron microscopy, flow cytometry, real-time quantitative PCR and Western blotting. Results Morphological changes of apoptotic JAR cells were observed under electron microscope. Flow cytometry showed that the number of apoptotic cells and necrotic cells in early stage were significantly higher than those in control group (P <0.01) . The expression of c-myc mRNA was 0.87 ± 0.12 and 0.45 ± 0.08 after treated with 20,40 mg.L-1 4-MS for 48 h, respectively. The expression of c-myc was 0.37 ± 0.02 and 0.25 ± 0.02, both lower than Control group (P <0.01). Conclusion 4-MS induced JAR cell apoptosis, the mechanism of which is related to the decrease of c-myc expression.
