淋巴细胞功能相关抗原-1(lymphocyte function-associated antigen 1,LFA-1)是白细胞上的重要粘附分子。本研究旨在探讨LFA-1缺失(LFA-1~(-/-))后小鼠血液、骨髓、脾脏中血液细胞分类和T细胞亚群的变化。本研究采用LFA-1~(-/-)小鼠转基因小鼠,经PCR鉴定其基因型;采用全自动血液检测仪测定血液细胞分类。为进一步了解淋巴细胞亚群的变化,采用流式细胞仪检测LFA-1~(-/-)和对照组小鼠血液、骨髓、和脾脏中T淋巴细胞亚群。结果表明,LFA-1~(-/-)小鼠血液中白细胞总数增多;在骨髓,血液和脾脏中CD3+T淋巴细胞增多。根据研究结果推测LFA-1可能调控CD3+细胞的分化。
Lymphocyte function-associated antigen 1 (LFA-1) is an important adhesion molecule on leukocytes. This study aimed to investigate the changes of blood cell class and T lymphocyte subsets in blood, bone marrow and spleen of mice after LFA-1 deficiency (LFA-1 ~ (- / -) -). In this study, LFA-1 ~ (- / -) mice transgenic mice were used to identify their genotypes by PCR. The blood cell classification was determined by automatic blood analyzer. To further understand the changes of lymphocyte subsets, T lymphocyte subsets in blood, bone marrow and spleen of LFA-1 ~ (- / -) and control mice were detected by flow cytometry. The results showed that the total number of white blood cells in LFA-1 ~ (- / -) mice increased; CD3 + T lymphocytes increased in bone marrow, blood and spleen. According to the results of the study speculated that LFA-1 may regulate CD3 + cell differentiation.
