目的:了解异丙酚是否通过影响大鼠不同脑区突触体质膜Ca~(2+)-ATP酶活性而产生中枢抑制效应。方法:SD大鼠30只,随机分为3组,分别腹腔注射异丙酚50mg·kg~(-1),100mg·kg~(-1)和生理盐水10ml·kg~(-1),待翻正反射消失后,立即断头,0℃~4℃下取脑,分区,匀浆,离心制备粗突触体,用钼蓝分光光度法测定Ca~(2+)-ATP酶活性。结果:腹腔注射异丙酚100mg·kg~(-1)能明显抑制大脑皮层,海马,脑干突触体质膜Ca~(2+)-ATP酶活性(尸<0.01)。结论:异丙酚对中枢神经系统的抑制作用,可能与其抑制脑突触体质膜Ca~(2+)-ATP酶活性有关。
AIM: To investigate whether propofol exerted a central inhibitory effect on Ca (2 +) - ATPase activity of synaptic plasma membrane in different brain regions in rats. Methods: Thirty SD rats were randomly divided into 3 groups. The rats were injected intraperitoneally with 50 mg · kg -1 and 100 mg · kg -1 of propofol and 10 ml · kg -1 of normal saline respectively. Right after the reflex disappeared, the decapitated head was immediately removed. The synaptosomes were harvested at 0 ℃ ~ 4 ℃, and the synaptosomes were prepared by centrifugation. The activity of Ca ~ (2 +) - ATPase was determined by molybdenum blue spectrophotometry. RESULTS: Intraperitoneal injection of propofol 100 mg · kg -1 could significantly inhibit the Ca ~ (2 +) - ATPase activity in the synaptic plasma membrane of the cerebral cortex, hippocampus and brainstem (P <0.01). CONCLUSION: Propofol inhibits the central nervous system and may be related to the inhibition of Ca ~ (2 +) - ATPase activity in the plasma membrane of synaptic bodies.
