目的:研究制备叶绿素亚铁脂质体的方法及产品缓释性。方法:采用硫酸铵梯度法制备叶绿素亚铁脂质体,以包封率为评价指标,在单因素实验的基础上,采用正交实验确定优化工艺,考察所得脂质体的缓释性。结果:叶绿素亚铁脂质体的最佳工艺条件为:叶绿素亚铁与磷脂的比例为1∶10、胆固醇与磷脂的质量比1∶2、孵育温度40℃、硫酸铵浓度为0.2mol/L。最佳工艺条件下制得3批脂质体的平均包封率为79.5%,脂质体中芯材在PBS溶液(pH7.2)中前3h的累积释放量低于15%。结论:实验中所选参实因子对叶绿素亚铁脂质体的包封率影响显著,其影响次序为药脂比>胆固醇与磷脂的比例>孵育温度>硫酸铵浓度,脂质体具有明显的缓释效果。
Objective: To study the method of preparation of chlorophyll ferrous liposomes and their sustained release. Methods: The ferrous chlorophyll was prepared by ammonium sulfate gradient method. The entrapment efficiency was used as the evaluation index. Based on the single factor experiment, the orthogonal experiment was used to determine the optimization process and the sustained release properties of the obtained liposomes were investigated. Results: The optimal conditions of ferrous chlorophyll were as follows: the ratio of ferrous chlorophyll to phospholipid was 1:10, the mass ratio of cholesterol to phospholipid was 1: 2, the incubation temperature was 40 ℃, the concentration of ammonium sulfate was 0.2 mol / L . The average entrapment efficiency of three batches of liposomes was 79.5% under the optimum conditions. The cumulative release of liposomes in PBS solution (pH7.2) was less than 15% in the first three hours. CONCLUSIONS: The factors selected in the experiment affect the entrapment efficiency of chlorophyll ferrous liposomes significantly, and the order of their effects is: ratio of lipid to lipid> ratio of cholesterol to phospholipid> incubation temperature> ammonium sulfate concentration, and liposomes have significant Slow release effect.
