目的探讨慢性脑血流灌注不足对老龄大鼠海马组织CA1区ABCA1的表达及血清中总胆固醇(Total cholesterod,TC)和高密度脂蛋白(High density lipoprotein,HDL)含量的影响。方法采用持久性双侧颈总动脉结扎法(Permanent occlusion of bilateral common carotid arteries,2VO)致老龄大鼠脑血流灌注不足,术后不同时间采用免疫组化法检测大鼠海马组织CA1区ABCA1的表达,全自动生化分析仪检测大鼠血清中HDL和TC的水平。结果术后1周,大鼠海马组织CA1区ABCA1表达阳性的细胞数与假手术组相比明显减少(P<0.01);术后2周,ABCA1阳性细胞数逐渐增多;术后3周,ABCA1阳性细胞数明显增加,数量最多,分布最广(P<0.01);术后4周,ABCA1阳性细胞数又开始下降(P<0.01)。术后2周,与假手术组相比,大鼠血清中HDL和TC含量显著升高(P<0.05),随着缺血时间的延长,又逐渐降低,术后4周与术后2周相比,差异有统计学意义(P<0.05)。结论 ABCA1和HDL、TC在血管性痴呆模型大鼠海马和血清中呈动态表达,推测ABCA1在血管性痴呆中发挥了保护作用。
Objective To investigate the effect of chronic cerebral hypoperfusion on the expression of ABCA1 and the levels of total cholesterin (TC) and high density lipoprotein (HDL) in hippocampus CA1 of aged rats. Methods Cerebral perfusion was induced in aged rats by permanent occlusion of bilateral common carotid artery occlusion (2VO). The expression of ABCA1 in CA1 of hippocampus was detected by immunohistochemistry at different time points after operation. Expression, automatic biochemical analyzer detection of rat serum HDL and TC levels. Results After 1 week, the number of ABCA1 positive cells in hippocampus CA1 of rats decreased significantly (P <0.01), and the number of ABCA1 positive cells increased gradually 2 weeks after operation. At 3 weeks after operation, ABCA1 The number of positive cells increased significantly, with the largest number and the most widely distributed (P <0.01). After 4 weeks, the number of ABCA1 positive cells began to decrease again (P <0.01). At 2 weeks after operation, HDL and TC contents in serum of rats were significantly increased (P <0.05) compared with those in sham-operation group, and gradually decreased with the prolongation of ischemic time. After 4 weeks and 2 weeks after operation Compared with the difference was statistically significant (P <0.05). Conclusion ABCA1, HDL and TC are dynamically expressed in the hippocampus and serum of vascular dementia model rats, suggesting that ABCA1 plays a protective role in vascular dementia.
