目的为配合3,4,5-三羟基苯甲酸(trihydroxybenzoic acid,TBA)脂质体的制备工艺研究,建立测定TBA脂质体中药物含量及包封率的高效液相色谱(HPLC)方法。方法超声薄膜法制备TBA脂质体,HPLC测定药物含量及包封率。色谱柱为AgilentTC-C18柱(4.6mm×150mm,5μm),流动相为甲醇-0.1%磷酸(15:85),流速为0.8mL.min-1,检测波长为273nm。结果 TBA在50~150mg.L-1内线性关系良好(r=0.9980,n=5),平均回收率为98.25%(RSD=0.45%)。结论超声薄膜法可以制备出包封率较高的TBA脂质体,HPLC测定脂质体的包封率,方法准确可靠,简单快速。
OBJECTIVE To establish a method for the preparation of 3,4,5-trihydroxybenzoic acid (TBA) liposomes, a high performance liquid chromatography (HPLC) method for the determination of drug content and entrapment efficiency in TBA liposomes was established. Methods TBA liposomes were prepared by ultrasonic membrane method. The drug content and entrapment efficiency were determined by HPLC. The column was an AgilentTC-C18 column (4.6 mm × 150 mm, 5 μm). The mobile phase consisted of methanol-0.1% phosphoric acid (15:85) at a flow rate of 0.8 mL.min-1 with a detection wavelength of 273 nm. Results TBA had a good linear relationship (r = 0.9980, n = 5) in the range of 50-150 mg.L-1 with an average recovery of 98.25% (RSD = 0.45%). Conclusion Ultrasound membrane method can prepare encapsulated TBA liposomes with high encapsulation efficiency. The encapsulation efficiency of liposomes is determined by HPLC. The method is accurate, reliable, simple and rapid.
