目的研究血管外膜依赖性舒张因子(ADRF)是否存在种属特异性和组织依赖性;对浴槽中分离的蛋白质进行初步研究。方法测定保存外膜和去外膜的胸主动脉环张力;SDS-PAGE电泳。结果①在Sprague-Dawley大鼠,与外膜完整的胸主动脉环相比,缺少外膜脂肪的动脉环对苯肾上腺素的量效曲线非平行左移。提示脂肪组织能够抑制苯肾上腺素诱导的胸主动脉环收缩。该现象能够在Wistar大鼠,豚鼠,家兔的胸主动脉发现;②而且取大网膜脂肪组织放入浴槽中能够减少苯肾上腺素诱导的收缩③ADRF的释放能够被染料木黄酮(亦名金雀异黄素,酪氨酸激酶抑制剂,10μmol.L-1)强烈抑制。但是金雀异黄素不能减弱已经分泌的ADRF的舒张血管作用;④从浴槽内液体中分离到5个蛋白质条带,分子量依次是74.0,59.8,54.4,28.7和13.8 ku。结论①血管外膜依赖性舒张因子(ADRF)是一种非种属特异性和不限于血管外膜脂肪组织分泌的蛋白质因子;②ADRF的全称应该从“adventitium-derived relaxing factor”修改为“adipocyte-derived relaxing factor”脂肪细胞依赖性舒张因子;③从浴槽内液体中分离到5个蛋白质条带。
Objective To investigate the existence of species-specific and tissue-dependent vascular adventitial relaxing factor (ADRF). To study the protein isolated from the bath. Methods The tension of the thoracic aorta of the preserved epicardium and epicardium was measured. SDS-PAGE electrophoresis was performed. RESULTS ① In Sprague-Dawley rats, the dose-response curves of phenylephrine in the arterial rings lacking the adventitia were non-parallel to the left aorta compared with intact thoracic aorta rings. Tip adipose tissue can inhibit phenylephrine-induced thoracic aortic ring contraction. This phenomenon can be found in the thoracic aorta of Wistar rats, guinea pigs and rabbits; ②and taking omental adipose tissue into the bath can reduce the phenylephrine-induced contraction ③ ADRF release can be genistein (also known as gold Isoflavin, tyrosine kinase inhibitor, 10 μmol.L-1) strongly inhibited. But genistein failed to attenuate the vasodilator effect of the secreted ADRF. (4) Five protein bands were separated from the liquid in the bath, and the molecular weights were 74.0, 59.8, 54.4, 28.7 and 13.8 ku, respectively. Conclusion ① Adventitial diastolic factor (ADRF) is a non-species-specific and not limited to the secretion of extravascular adventitial protein factor; ② The full name of ADRF should be modified from “adventitium-derived relaxing factor” to “adipocyte- derived relaxing factor ”adipocyte-dependent relaxation factor; ③ isolated from the bath liquid five protein bands.
