为进一步揭示显性核不育亚麻不育分子机理,以显性核不育亚麻不育株分离的可育花蕾和不育花蕾为研究材料,采用实时定量PCR技术和原位杂交技术,开展了G-E07-830、G-E07-330和G-E07-100这3个花蕾发育相关基因在亚麻花蕾中的时空表达模式。结果表明:原位杂交中G-E07-830、G-E07-330和G-E07-100基因在亚麻可育和不育花蕾绒毡层中表达特异性信号;实时定量PCR得出在早期不育亚麻花蕾中表达量显著大于其他发育时期的花蕾。
In order to further reveal molecular mechanism of flax infertility in dominant genic male sterility, fertile flower buds and sterile flower buds isolated from male sterile flax lines were used as experimental materials. Real-time PCR and in situ hybridization G-E07-830, G-E07-330 and G-E07-100 bud-related gene development in flax flower buds in the spatial and temporal expression patterns. The results showed that G-E07-830, G-E07-330 and G-E07-100 genes were expressed in tapetumine and sterile tapetum in situ hybridization. Real-time quantitative PCR showed that G-E07-830, G- The expression levels in the flax buds were significantly greater than those in other developmental stages.
