目的评价3′-脱氧-3′-18F-氟代胸苷(18F-FLT)作为一种新的PET示踪剂在恶性肿瘤诊断中的作用。方法采用荷肺腺癌T739小鼠模型。实验组经尾静脉注入18F-FLT后30、60、90、120min(每时间点8只小鼠)分别用井型探测仪测量18F-FLT的生物分布,并行PET显像。对照组(n=8)小鼠在注入18F-FDG后60min测量生物分布并行PET显像。结果18F-FLT在肿瘤部位有相当高的放射性摄取,在肾脏和脾脏亦有很高的放射性,肿瘤对血、肌肉及肺的T/NT比值均大于2。应用18F-FLT的肿瘤PET显像清晰。结论肺腺癌组织中18F-FLT摄取高于正常组织,可通过PET清晰显像,为进一步的临床研究提供了依据。
Objective To evaluate the role of 3’-deoxy-3’-18F-fluorothymidine (18F-FLT) as a new PET tracer in the diagnosis of malignant tumors. Methods Lung adenocarcinoma T739 mouse model was used. The experimental group was injected with 18F-FLT through the tail vein 30,60,90,120 min (8 mice per time point) were measured by well-type detector 18F-FLT biodistribution, parallel PET imaging. Biodistribution parallel PET imaging was measured in control (n = 8) mice at 60 min after injection of 18F-FDG. Results 18F-FLT had a high radioactive uptake in the tumor site and high radioactivity in the kidney and spleen. The T / NT ratio of tumor to blood, muscle and lung was greater than 2. Tumors PET imaging with 18F-FLT was clear. Conclusion The uptake of 18F-FLT in lung adenocarcinoma tissues is higher than that of normal tissues, which can be clearly visualized by PET, which provides the basis for further clinical research.
