目的探讨清脂护肝方对非酒精性脂肪肝大鼠肝组织病理变化及纤维化指标的影响。方法 76只SD大鼠除正常组10只外,其余大鼠以高脂饲料(88%普通饲料+2%胆固醇+10%猪油)喂养造模,12周后验证非酒精性脂肪肝大鼠造模成功。将造模成功的大鼠再随机分为模型组、阳性对照组、中药低剂量组、中药中剂量组和中药高剂量组,各组给予相应干预措施共6周。实验结束后进行大鼠肝组织半定量分析和光电镜下病理学观察,并检测大鼠肝纤维化指标,包括血清IV型胶原(CIV)、层粘连蛋白(LN)及肝组织匀浆CIV。结果与模型组比较,清脂护肝方各剂量组肝组织匀浆CIV、血清LN明显降低,差异有统计学意义(P<0.05,P<0.01)。与模型组比较,清脂护肝方各剂量组脂肪变性程度明显减轻,差异有统计学意义(P<0.01)。清脂护肝方中剂量组、高剂量组脂肪变性积分较阳性治疗组明显降低,差异有统计学意义(P<0.05,P<0.01)。结论清脂护肝方具有良好的抗纤维化及脂肪变性作用,能改善实验性脂肪肝大鼠肝组织病理变化。
Objective To investigate the effect of Qingzhi Hugan Decoction on pathological changes of liver tissue and fibrosis in nonalcoholic fatty liver disease rats. Methods Totally 76 SD rats were fed with high fat diet (88% normal diet + 2% cholesterol + 10% lard) except 10 rats in normal group. After 12 weeks, nonalcoholic fatty liver rats Modeling success. The successful rats were randomly divided into model group, positive control group, low-dose traditional Chinese medicine group, medium-dose traditional Chinese medicine group and high-dose traditional Chinese medicine group. Corresponding interventions were given in each group for 6 weeks. After the experiment, semi-quantitative analysis of liver tissues and pathological observation under light microscope were performed. The indexes of liver fibrosis in rats were detected, including serum type IV collagen (CIV), laminin (LN) and liver homogenate CIV. Results Compared with the model group, the CIV and the serum level of LN in each group of Qingzhi Hugan Decoction were significantly decreased (P <0.05, P <0.01). Compared with the model group, the degree of fatty degeneration of each dose group of Qingzhi Hugan decoction was significantly reduced (P <0.01). Compared with the positive group, the middle-dose Qingzhi Hugan formula group and the high dose group had significantly lower scores of steatosis, the difference was statistically significant (P <0.05, P <0.01). Conclusion Qingzhi Hugan Decoction has good anti-fibrosis and steatosis, and can improve the pathological changes of liver in experimental fatty liver rats.
