为了研究橘核中柠檬苦素超声提取工艺,试验采用四因素三水平的正交试验设计,并对最佳提取工艺进行稳定性试验,用3种方法比较6种不同产地橘核中柠檬苦素的含量。结果表明:乙醇浓度为75%、液料比为20∶1、提取时间为30 min、超声功率为500 w,提取效果最好,且稳定可行;在此条件下提取,广东产的橘核中柠檬苦素的含量最高,山东产的最低;与索氏法、回流法比较,超声法能明显提高橘核柠檬苦素的提取含量,说明此最佳工艺可提高药材的利用率。
In order to study the ultrasonic extraction process of limonin in orange nucleus, four factors and three levels of orthogonal design were used in the experiment. The best extraction process was tested for stability. Three methods were used to compare limonin Content. The results showed that the optimum extraction conditions were 75% ethanol concentration, 20:1 liquid-to-liquid ratio, 30 min extraction time and 500 w ultrasonic power. The extraction was the best and stable. Under these conditions, The content of limonin is the highest and the lowest in Shandong. Compared with Soxhlet method and refluxing method, the ultrasonic method can obviously improve the content of limonin extracted from the orange nucleus, indicating that the best technique can improve the utilization rate of medicinal materials.
