目的探讨凝血酶、胰蛋白酶和类胰蛋白酶等丝氨酸蛋白酶对肥大细胞P815分泌IL-4的影响。方法肥大细胞培养后用凝血酶、胰蛋白酶、类胰蛋白酶单独或与相应抑制剂凝血酶抑制剂、大豆胰蛋白酶抑制剂、α抗胰蛋白酶抑制剂、亮肽素结合处理肥大细胞,收集上清并用ELISA方法检测IL-4的水平。结果胰蛋白酶、类胰蛋白酶以浓度依赖的方式促进肥大细胞P815分泌IL-4。大豆胰蛋白酶抑制剂和α抗胰蛋白酶能阻断胰蛋白酶引起的肥大细胞IL-4分泌(P<0.05)。亮肽素能阻断类胰蛋白酶引起的肥大细胞IL-4分泌(P<0.05)。而凝血酶对肥大细胞IL-4的分泌功能无影响。结论胰蛋白酶和类胰蛋白酶刺激肥大细胞IL-4分泌。
Objective To investigate the effect of serine protease such as thrombin, trypsin and tryptase on the secretion of IL-4 by mast cell P815. Methods After mast cells were cultured, mast cells were treated with thrombin, trypsin, tryptase alone or in combination with the corresponding inhibitors of thrombin inhibitor, soybean trypsin inhibitor, α antitrypsin inhibitor and leupeptin, and the supernatant was collected The level of IL-4 was detected by ELISA. Results Trypsin and tryptase promoted the secretion of IL-4 by mast cell P815 in a concentration-dependent manner. Soybean trypsin inhibitor and alpha antitrypsin can block trypsin-induced mast cell IL-4 secretion (P <0.05). Leupeptin blocked the tryptase-induced mast cell IL-4 secretion (P <0.05). Thrombin has no effect on the secretion of IL-4 in mast cells. Conclusion Trypsin and tryptase stimulate the secretion of IL-4 in mast cells.
