目的检测Wnt信号通路成分β-连环蛋白(β-catenin)、细胞周期蛋白D1(cyclinD1)在卵巢浆液性腺癌中的表达情况,探讨两者的关系及与肿瘤FIGO分期、病理分级、患者预后的关系。方法应用免疫组织化学Power Vision(PV)二步法检测60例卵巢浆液性腺癌中β-catenin、cyclinD1的表达,并运用χ2检验分析其与肿瘤FIGO分期、病理分级的关系,运用Kaplan-Meier法分析其与预后的关系,运用Spearman法分析两者的相关性。结果 (1)β-catenin的表达与FIGO分期相关,Ⅲ、Ⅳ期组的阳性表达高于Ⅰ、Ⅱ期组(P=0.003);与病理分级无关(P=0.817);与患者预后相关,β-catenin阳性组的生存率低于阴性组(P=0.034),晚期(Ⅲ、Ⅳ期)患者中β-catenin与预后无关(P=0.495)。(2)cyclinD1的表达与FIGO分期无关(P=0.829);与病理分级相关,高级别组的阳性表达高于低级别组(P=0.017);与患者预后相关,cyclinD1阳性组的生存率低于阴性组(P=0.009),晚期(Ⅲ、Ⅳ期)患者中cyclinD1与预后相关(P=0.039)。(3)cyclinD1与β-catenin的表达呈正相关(P=0.014),两者的共同阳性率为56.67%,共同阴性率为13.33%。结论 Wnt信号通路成分β-catenin、cyclinD1可能在卵巢浆液性腺癌的发生发展中发挥了作用,两者高表达提示预后不良,检测其表达情况有望指导临床工作。
Objective To detect the expression of β-catenin and cyclinD1 in ovarian serous adenocarcinoma, and to explore their relationship with the FIGO stage, pathological grade, prognosis of patients relationship. Methods The expressions of β-catenin and cyclinD1 in 60 cases of ovarian serous adenocarcinoma were detected by immunohistochemistry Power Vision (PV) two-step method. The relationship between the expression of β-catenin and cyclinD1 in FIGO stage and pathological grade was analyzed by Chi-square test. Kaplan-Meier method Analyze its relationship with prognosis, using Spearman analysis of the correlation between the two. Results (1) The expression of β-catenin was correlated with FIGO staging. The positive expression of β-catenin in stage Ⅲ and Ⅳ was higher than that in stage Ⅰ and Ⅱ (P = 0.003), but not with the pathological grade (P = 0.817) The survival rate of β-catenin positive group was lower than that of negative group (P = 0.034). The β-catenin was not related to prognosis in advanced stage Ⅲ and Ⅳ (P = 0.495). (2) The expression of cyclinD1 was not correlated with FIGO stage (P = 0.829). The positive expression of cyclinD1 in high grade group was higher than that in lower grade group (P = 0.017). The prognosis of patients with cyclinD1 positive group was lower than that of low grade group CyclinD1 was associated with prognosis in the negative (P = 0.009) and advanced (III, IV) patients (P = 0.039). (3) There was a positive correlation between the expression of cyclinD1 and β-catenin (P = 0.014). The positive rate of cyclinD1 was 56.67% and the common negative rate was 13.33%. Conclusions β-catenin and cyclinD1, a component of Wnt signaling pathway, may play a role in the development of ovarian serous adenocarcinoma. The high expression of both of them may lead to poor prognosis. Detecting the expression of β-catenin and cyclinD1 may lead to clinical work.
