本文报道胃复安在兔体内的药代动力学。按较大剂量(20mg·kg~(-1))快速静注后,血药浓度数据采用自编非线性算法和程序在Apple——Ⅱ微机上作曲线拟合后,依据最小残差平方和(Re)和AIC值作模型识别。5只兔均按二室模型拟合较为合适。算得参数:t_1/2α和t_1/2日分别为4.6±3.7和75.3±15.2min;Vc、Vd(area)和Vd(ss)分别为2.98±1.80、4.94±0.65和4.72±0.51L·kg~(-1);Cl为46.6±10.5ml·min.kg~(-1)。作统计矩分析,AUC、MRT和VRT分别为446±93μg·hr·ml~(-1)、104.5±19.6min和12062±4469min~2。
This article reports the pharmacokinetics of metoclopramide in rabbits. After bolus injection of 20 mg · kg ~ (-1) bolus, the blood plasma concentration data were fit to the curve of Apple - Ⅱ computer with self - (Re) and AIC values for model identification. Five rabbits were fitted by two-compartment model is more appropriate. The calculated parameters were: 4.6 ± 3.7 and 75.3 ± 15.2 min for t 1 / 2α and t 1/2, and 2.98 ± 1.80, 4.94 ± 0.65 and 4.72 ± 0.51L · kg -1 for Vc, Vd (area) and Vd (-1); Cl was 46.6 ± 10.5ml · min.kg -1. For statistical moment analysis, AUC, MRT and VRT were 446 ± 93 μg · hr · ml -1, 104.5 ± 19.6 min and 12062 ± 4469 min ~ 2, respectively.
