目的:建立RP-HPLC同时测定白鲜皮中白鲜碱、黄柏酮和梣酮含量的方法。方法:采用RPHPLC,70%乙醇超声提取,色谱条件:色谱柱Alltima C18(4.6 mm×250 mm,5μm),保护柱All-GuardTM C18(4.6 mm×7.5 mm,5μm),流动相乙腈-0.1%磷酸梯度洗脱,流速1.0 mL/min,进样量10μL,柱温25℃,检测波长238 nm。结果:该3种成分分离良好,线性范围依次为0.4~16.0,10~400和1.6~64.0 mg/L,加样回收率分别为99.95%,98.91%,100.02%,RSD分别为:1.81%,1.83%,2.62%。结论:本方法简单、快捷,重现性好,准确率高,可用于白鲜皮药材的质量控制。
OBJECTIVE: To establish a method for the simultaneous determination of white bastardine, berberine and choristone in fresh white peel by RP-HPLC. METHODS: The samples were extracted with RPHPLC and 70% ethanol. The chromatographic conditions were as follows: Alltima C18 column (4.6 mm × 250 mm, 5 μm), All-Guard ™ C18 column (4.6 mm × 7.5 mm, Phosphoric acid gradient elution, flow rate 1.0 mL / min, the injection volume 10μL, column temperature 25 ℃, detection wavelength 238 nm. Results: The three components were well separated and the linear range was 0.4 ~ 16.0, 10 ~ 400 and 1.6 ~ 64.0 mg / L, the recoveries were 99.95%, 98.91% and 100.02%, respectively. The RSDs were 1.81% 1.83%, 2.62%. Conclusion: The method is simple, rapid, reproducible, accurate and can be used for the quality control of the fresh white skin medicinal materials.
