目的 :研究诱导骨肉瘤特异性细胞毒T淋巴细胞 (OSS CTL) ,及其体内外的抗肿瘤作用。方法 :通过生化方法提取纯化骨肉瘤相关抗原 (OSAA) ,并与低剂量IL 2 、CD3 、McAb协同刺激骨肉瘤致敏的淋巴细胞诱导产生OSS CTL。结果 :OSS CTL表型特征为以CD3 +CD8+CTL为主的异质细胞群 ;对OSAA相关的骨肉瘤细胞显示高亲和杀伤活性。体内实验表明OSS CTL明显抑制裸鼠皮下骨肉瘤的生长和显著延长荷瘤裸鼠生存时间。结论 :OSS CTL可望成为骨肉瘤免疫治疗的新的免疫效应细胞。
Objective: To study the induction of osteosarcoma-specific cytotoxic T lymphocytes (OSS CTL) and its anti-tumor effect in vitro and in vivo. Methods: Osteosarcoma related antigen (OSAA) was purified by biochemical methods. Osteosarcoma - sensitized lymphocytes were synergistically stimulated with IL - 2, CD3 and McAb to induce OSS CTL. Results: The phenotypic characteristics of OSS CTL were heterogeneous cell population mainly including CD3 + CD8 + CTL. OSA-related osteosarcoma cells showed high affinity and cytotoxic activity. In vivo experiments showed that OSS CTL significantly inhibited the growth of subcutaneous osteosarcoma in nude mice and significantly prolonged the survival of tumor-bearing nude mice. Conclusion: OSS CTL is expected to become a new immune effector cells for immunotherapy of osteosarcoma.
