脱氧核糖核酸(DNA)碱基的测定在分子遗传学上有着十分重要的意义。近年来武者宗一郎、邓家祺等报导了核酸碱基的阳极溶出伏安法,检测下限为1×10~(-7)M。Temerk~ε用脉冲极谱法直接测定腺嘌呤,检测下限为1.5×10~(-6)~4.5×10~(-6)M。我们发现在0.5M抗坏血酸介质中,腺嘌呤在单扫描示波极谱仪上能产生灵敏的吸附还原波,检测下限为5×10~(-8)M。根据所拟方法测定了DNA样品中腺嘌呤,结果较好。本文还
Determination of DNA bases in molecular genetics has a very important significance. In recent years, Sobuchi Soichiro, Deng Jiaqi et al reported anodic stripping voltammetry of nucleic acid bases, the detection limit of 1 × 10 ~ (-7) M. The direct determination of adenine by Temerk ~ εpulsive polarographic method showed that the detection limit was 1.5 × 10 -6 -6.5 × 10 -6 M. We found that in 0.5M ascorbic acid medium, adenine can produce sensitive adsorption reduction wave on a single-scan oscilloscope with a detection limit of 5 × 10 -8 M.
