目的:探讨17β-雌二醇对抗癫痫药物短期应用导致的未成熟脑受体表达降低的保护作用。方法:3 d、7 d、14 d、21 d SD大鼠各7窝,共28窝,随机分为对照组、抗癫痫药物组及雌二醇组,抗癫痫药物组又分为苯巴比妥组、托吡酯组以及联合用药组,每组SD大鼠12只。HE染色了解海马CA1区结构,免疫组化标记海马CA1区谷氨酸受体(GluR1、NR2B),灰度分析检测受体表达情况。结果:各年龄段17β-雌二醇药物保护组较未使用17β-雌二醇保护的抗癫痫药物组GluR1、NR2B受体表达均增多(P<0.01);但各17β-雌二醇保护组受体表达较对照组仍有降低(P<0.01)。结论:17β-雌二醇对抗癫痫药物造成的脑损伤有保护作用,但保护作用尚有限。
Objective: To explore the protective effect of 17β-estradiol on the decrease of immature brain receptor expression induced by short-term application of antiepileptic drugs. Methods: Seven SD rats were randomly divided into control group, antiepileptic group and estradiol group. The antiepileptic group was divided into phenobarbital group Tuo group, topiramate group and combination group, 12 SD rats in each group. HE staining was used to detect the structure of hippocampal CA1 region, immunohistochemistry labeled glutamate receptors (GluR1, NR2B) in hippocampal CA1 region, gray scale analysis of receptor expression. Results: The expressions of GluR1 and NR2B receptors in 17β-estradiol-protected group were higher than those in 17β-estradiol-protected antiepileptic group (P <0.01) Receptor expression was still lower than the control group (P <0.01). Conclusion: 17β-estradiol has a protective effect on brain damage induced by antiepileptic drugs, but the protective effect is still limited.
