以紫薇为试材,利用MISA软件对转录组测序获得45 308条unigenes中的SSR特征进行分析。结果表明:9 074条unigenes存在10 905个SSR位点,1 486条unigenes含有2个以上的SSR位点,550个位点是复合重复位点;从7 620个SSRs位点中设计出4 501对SSR引物;随机选择68对SSR引物进行检测,其中61对引物可以成功的扩增出清晰可用的条带,28对引物有多态性。
Using crape mycelium as test material, the SSR characteristics of 45 308 unigenes were analyzed by MISA software. The results showed that there were 10 905 SSR loci in 9 074 unigenes, 1 486 unigenes contained more than 2 SSR loci and 550 loci were composite repeat loci. From 7 620 SSRs loci, 4 501 A total of 68 pairs of SSR primers were selected for SSR primers. Sixty-one pairs of SSR primers were successfully used to amplify clear bands and 28 pairs of primers were polymorphic.
