目的研究一种快速、简便、准确、稳定的化疗药敏检测方法。方法通过对现有MTT化学比色法的药物浓度,药物与瘤细胞的接触时间以及孵育和观测的方式进行精细地选择和比较。结果:①敏感的药物即使在1/100ppc时亦有较高的抑瘤率;不敏感的药物即使在10ppc时也只有较低的抑瘤率。②在药物接触4~12d小时期间,抑瘤率上升比较明显,12~48小时期间,抑瘤率变化比较平缓。③普通保温恒温箱与CO2孵箱孵育后的检测结果十分接近(P>0.05)。④活细胞上所形成的蓝紫色放射状结晶(formazan)可用倒镜及光镜进行观测。⑤敏感药物与非敏感药物的显色结果区别明显,可用肉眼进行观测。结论本法简便、快速、经济、准确、稳定,不需昂贵仪器设备,即可定量测定,也可半定量观测,便于在中小医院推广使用。
Objective To study a rapid, simple, accurate, and stable method for detecting susceptibility to chemotherapy. Methods The selection and comparison of the drug concentration of the existing MTT chemical colorimetry, the contact time of the drug with the tumor cells, and the manner of incubation and observation were carefully selected. RESULTS: 1 Sensitive drugs had higher tumor inhibition rates even at 1/100ppc; insensitive drugs had low tumor inhibition rates even at 10 ppc. 2 During the period of 4 to 12 days after exposure to the drug, the rate of inhibition of tumor growth increased significantly. During the period of 12 to 48 hours, the rate of inhibition of tumor growth was relatively flat. 3 The results of incubation after incubation with an ordinary incubation thermostat and a CO2 incubator were very close (P>0.05). 4 Formazan blue-violet crystals (formazan) formed on living cells can be observed with a mirror and a light microscope. 5 The chromogenic results of sensitive and non-sensitive drugs are significantly different and can be observed with the naked eye. Conclusion This method is simple, rapid, economical, accurate and stable. It can be quantitatively measured without expensive instruments and equipment. It can also be semi-quantitatively observed and can be easily used in small and medium-sized hospitals.
