黄瓜花叶病毒(Cucumber mosaic virus,CMV)是番茄病毒病的主要病原之一。为了研究CMV与番茄相互作用的分子机制,本研究以受CMV侵染的番茄叶片为材料,构建了番茄的cDNA文库。文库总库容量为2.346×106 cfu,其中0.5~1 kb片段文库库容量为1.02×106 cfu;1~2 kb片段文库库容量为8.56×105 cfu;大于2 kb片段文库库容量为4.7×105 cfu。三个子文库各检测20个克隆,重组率为100%。随机挑取30个克隆进行测序,获得29条有效序列,有23条序列包含完整的开放阅读框,表明获得的文库质量较高。高质量番茄cDNA文库的获得,为研究CMV-番茄相互作用奠定了基础。
Cucumber mosaic virus (CMV) is one of the major pathogens of tomato virus disease. In order to study the molecular mechanism of the interaction between CMV and tomato, we constructed a cDNA library of tomato using CMV infected tomato leaves as material. The library has a total library capacity of 2.346 × 106 cfu, of which 0.5-1 kb fragment library has a capacity of 1.02 × 106 cfu, a 1 ~ 2 kb fragment library has a capacity of 8.56 × 105 cfu, and a library with a size larger than 2 kb has a capacity of 4.7 × 105 cfu. Each of the three sub-libraries detected 20 clones with a recombination rate of 100%. Thirty randomly selected clones were sequenced and 29 validated sequences were obtained. Twenty-three of these sequences contained the complete open reading frame, indicating that the quality of the obtained library was high. The access of high quality tomato cDNA library laid the foundation for the study of CMV-tomato interaction.
