目的近年来,有关pre-miR-27a基因rs895819位点多态性与大肠癌易感性关系的研究日益增多,但结论并不一致。本研究从循证医学角度,综合评价pre-miR-27a基因rs895819位点多态性与大肠癌易感性的关系。方法通过检索数据库PubMed、Cochrane Library、EMBASE以及中国知网数据库、万方数据库,收集有关pre-miR-27a基因rs895819位点多态性与大肠癌罹患风险关系的病例-对照研究。检索时间1998-01-01-2015-11-01。依据文献纳入及排除标准筛选相关文献,提取基本数据信息并进行文献质量评估。采用Stata 12.0软件行Meta分析,计算合并OR值和95%CI,并进一步行亚组分析和敏感性分析。结果最终纳入6个病例-对照研究(包括2 025例大肠癌患者和2 320例非肿瘤对照者)。Meta分析结果显示,pre-miR-27a基因rs895819多态性与大肠癌罹患风险具有显著的相关性,Gvs A:OR=1.18,95%CI=1.08~1.30;GG vs AA/AG:OR=1.52,95%CI=1.26~1.97;GG vs AA:OR=1.53,95%CI=1.26~1.86,P值均<0.05。亚组分析结果发现,亚洲人群也有相似的结论,Gvs A:OR=1.21,95%CI=1.09~1.35;AG/GGvs AA:OR=1.15,95%CI=1.00~1.32;GG vs AA/AG:OR=1.57,95%CI=1.27~1.93;GG vs AA:OR=1.59,95%CI=1.27~1.98,P值均<0.05。结论 pre-miR-27a基因rs895819多态性与大肠癌易感性之间具有相关性,并且GG基因型具有增加罹患大肠癌的风险。
Purpose In recent years, there is an increasing number of studies on the relationship between rs895819 polymorphism of pre-miR-27a gene and susceptibility to colorectal cancer, but the conclusions are not consistent. This study from the perspective of evidence-based medicine, a comprehensive evaluation of pre-miR-27a gene rs895819 polymorphism and the relationship between colorectal cancer susceptibility. Methods A case-control study on the relationship between the polymorphism of rs895819 site of pre-miR-27a gene and the risk of colorectal cancer was collected by searching database PubMed, Cochrane Library, EMBASE, CNKI and Wanfang database. Retrieval time 1998-01-01-2015-11-01. According to the literature inclusion and exclusion criteria screening related documents, extract the basic data and information quality assessment. Meta-analysis was performed with Stata 12.0 software to calculate the combined OR and 95% CI, and further subgroup analysis and sensitivity analysis. Results Six case-control studies (including 2 025 cases of colorectal cancer and 2 320 non-tumor controls) were included. Meta analysis showed that there was a significant correlation between rs895819 polymorphism of pre-miR-27a gene and the risk of colorectal cancer. Gvs A: OR = 1.18, 95% CI = 1.08-1.30; GG vs AA / AG: OR = 1.52 , 95% CI = 1.26-1.97; GG vs AA: OR = 1.53, 95% CI = 1.26-1.86, all P <0.05. The subgroup analysis found that the Asian population also had similar conclusions: Gvs A: OR = 1.21, 95% CI = 1.09-1.35; AG / GGvs AA: OR = 1.15, 95% CI = 1.00-1.32; : OR = 1.57, 95% CI = 1.27-1.93; GG vs. AA: OR = 1.59, 95% CI = 1.27-1.98, P <0.05. Conclusion The rs895819 polymorphism of pre-miR-27a gene is associated with the susceptibility to colorectal cancer, and the GG genotype may increase the risk of developing colorectal cancer.
