目的研究靶向METCAM/MUC18的siRNA在肝癌细胞迁移、侵袭中的作用。方法培养肝癌细胞株Hep G2并转染MUC18的siRNA和阴性对照(NC)的siRNA,转染siRNA后检测细胞的迁移和侵袭数目、MMPs及上皮间质转化标志基因的m RNA表达量。结果 MUC18-siRNA组Hep G2细胞中MUC18的m RNA表达量显著低于NC-siRNA组;转染siRNA后12 h、24 h,MUC18-siRNA组Hep G2细胞的迁移数目和侵袭数目均显著低于NC-siRNA组;转染siRNA后24 h,MUC18-siRNA组Hep G2细胞MMP8、MMP9、MMP14、N-cadherin、Vimentin的m RNA表达量均显著低于NC-siRNA组,E-cadherin的m RNA表达量显著高于NC-siRNA组。结论靶向METCAM/MUC18的siRNA能够通过抑制MMPs表达和上皮间质转化来抑制肝癌细胞的迁移、侵袭过程。
Objective To study the role of siRNA targeted to METCAM / MUC18 in the migration and invasion of hepatoma cells. Methods Hep G2 cells were transfected with siRNA of MUC18 and negative control (NC). The expression of m RNA in the cells was detected by flow cytometry. Results MUC18 mRNA expression in MUC18-siRNA group was significantly lower than that in NC-siRNA group. The number of MUC18-siRNA group and the number of invasion in MUC18-siRNA group were significantly lower than those in MUC18-siRNA group NC-siRNA group. The mRNA expression of MMP8, MMP9, MMP14, N-cadherin and Vimentin in Hep G2 cells of MUC18-siRNA group was significantly lower than that of NC-siRNA group 24 h after siRNA transfection The expression level was significantly higher than NC-siRNA group. Conclusions siRNA targeting METCAM / MUC18 can inhibit the migration and invasion of hepatoma cells by inhibiting the expression of MMPs and transforming epithelial cells.
