                                目的观察五子衍宗丸对肾精亏虚证大鼠精液质量及睾丸支持细胞波形蛋白(vimentin)表达的影响。方法 SD雄性大鼠随机分为5组,即空白组,模型组,五子衍宗丸低、中、高剂量组,每组5只。空白组每天给予5%羧甲基纤维素钠(CMC-Na)灌胃,模型组和五子衍宗丸组每天上午都给予雷公藤多甙(GTW)20mg/kg灌胃,下午模型组给予5%CMC-Na 50mg/kg,五子衍宗丸低、中、高剂量组分别给予五子衍宗丸(CMC-Na)0.5kg、1.0kg、2.0g/kg灌胃30 d,第31天取材,通过计算机辅助精子分析系统(CASA)检测精液质量,Western Blot法检测睾丸支持细胞vimentin表达。结果与空白组相比,肾精亏虚证大鼠精子浓度(12.38×10~6/mL vs 45.20×10~6/mL)、精子总数(32.87×10~6 vs 98.09×10~6)、前向运动(PR)精子百分率(30.40%vs 59.00%)、总活力(46.60%vs 87.60%)均显著降低(P<0.05),vimentin含量明显降低(0.29 vs 0.43,P<0.05);与模型组相比,五子衍宗丸高剂量组精子浓度(36.15×10~6/mL)、精子总数(85.44×10~6)、PR(57.50%)、总活力(77.75%)及vimentin含量(0.41)均显著提高(P<0.05),五子衍宗丸中剂量组精子总数(73.48×10~6)、PR(54.00%)、总活力(69.75%)显著提高(P<0.01)。结论五子衍宗丸可以改善生精功能,机制可能是修复支持细胞骨架。
Objective To observe the effect of Wuzi Yanzong Pills on sperm quality and vimentin expression in testis supporting cells in rats with deficiency of kidney essence. Methods SD male rats were randomly divided into five groups: blank group, model group and WZKY low, medium and high dose groups, with 5 rats in each group. Rats in the model group and Wuzi Yanzong Pill group were given GTW 20mg / kg orally every morning, while the model group was given 5% sodium carboxymethylcellulose (CMC-Na) 0.5mg, 1.0kg and 2.0g / kg of Wuzi Yanzong Pills (CMC-Na) were administered intragastrically for 30 days after administration of CMC-Na 50mg / kg and Wuziyanzong pill low, medium and high dose groups, respectively. Sperm quality was measured by Sperm Analysis System (CASA), and vimentin expression was detected by Western Blot. Results Compared with the blank group, the sperm concentration (12.38 × 10 ~ 6 / mL vs 45.20 × 10 ~ 6 / mL), the total number of sperm (32.87 × 10 ~ 6 vs 98.09 × 10 ~ 6) The percentage of preeclamptic motility (PR) sperm (30.40% vs 59.00%) and total vitality (46.60% vs 87.60%) were significantly lower than that of model Sperm concentration (36.15 × 10 ~ 6 / mL), sperm count (85.44 × 10 ~ 6), PR (57.50%), total activity (77.75%) and vimentin content (0.41) (P <0.05). The total sperm count (73.48 × 10 ~ 6), PR (54.00%) and total activity (69.75%) of Wuzi Yanzong Pill medium dose group were significantly increased (P <0.01). Conclusion Wuzi Yanzong pill can improve the function of spermatogenesis, the mechanism may be to repair the supporting cytoskeleton.
