[目的]探讨携带内皮抑素基因的腺病毒(pLP-Ad-ES)对荷人卵巢癌裸鼠移植瘤的抑制作用及其作用机制。[方法]采用细胞培养、移植瘤模型建立、RT-PCR、免疫组化等方法观察pLP-Ad-ES、pLP-Ad-ES+顺铂(DDP)对裸鼠移植瘤的影响及VEGF、MMP-2、Endostatin的表达。[结果]pLP-Ad-ES、pLP-Ad-ES+DDP能够抑制裸鼠移植瘤的生长,有明显的抑瘤作用(P<0.05);经腺病毒作用后,裸鼠移植瘤中MMP-2、VEGF呈现低表达、Endostatin呈现高表达。[结论]pLP-Ad-ES能够抑制卵巢癌裸鼠移植瘤的生长;同时能够抑制VEGF、MMP-2的表达,增强Endostatin的表达,提示内皮抑素的作用机制可能与抑制血管生成和调节基质金属蛋白酶有关。
[Objective] To investigate the inhibitory effect of adenovirus carrying endostatin gene (pLP-Ad-ES) on human ovarian carcinoma xenografts in nude mice and its mechanism. [Methods] The effect of pLP-Ad-ES, pLP-Ad-ES + cisplatin (DDP) on xenografts in nude mice and the effect of VEGF, MMP- 2, Endostatin expression. [Results] pLP-Ad-ES and pLP-Ad-ES + DDP could inhibit the growth of xenografted tumor in nude mice and had obvious antitumor activity (P <0.05). After adenovirus- 2, VEGF showed low expression, Endostatin showed high expression. [Conclusion] pLP-Ad-ES can inhibit the growth of transplanted ovarian cancer xenografts in nude mice, meanwhile inhibit the expression of VEGF and MMP-2 and enhance the expression of Endostatin, suggesting that the mechanism of endostatin may be related to inhibiting angiogenesis and regulating matrix Metalloproteinase related.
