目的:观察高剂量丹参注射液静脉注射对小鼠肺组织的损伤作用,探讨临床高剂量使用丹参注射液的安全性,为其临床用药提供参考。方法:将30只小鼠随机分为对照组、低剂量丹参注射液组及高剂量丹参注射液组,分别每日尾静脉注射生理盐水、低剂量丹参注射液(0.64 g/kg)及高剂量丹参注射液(5.76 g/kg)。连续注射5 d后取小鼠肺组织进行HE染色;采用ELISA法检测外周血血清中肿瘤坏死因子-α(TNF-α)与白介素-1β(IL-1β)的浓度。结果:正常组与低剂量组小鼠肺组织未见异常。高剂量丹参注射液组小鼠肺组织呈弥漫性出血,并伴炎性细胞浸润;与正常组及低剂量丹参注射液组比较,高剂量丹参注射液组TNF-α和IL-1β的浓度显著增高(P<0.05)。结论:高剂量丹参注射液可导致小鼠肺组织损伤,诱导炎性反应。
Objective: To observe the damaging effect of intravenous injection of high dose Danshen injection on the lung tissue of mice and to explore the safety of high dose Danshen injection and to provide a reference for its clinical use. Methods: Thirty mice were randomly divided into control group, low dose Salviae Miltiorrhizae injection group and high dose Salviae Miltiorrhizae injection group, and received daily intravenous injection of normal saline, low dose Salvia miltiorrhiza injection (0.64 g / kg) and high dose Salvia injection (5.76 g / kg). After 5 days of continuous injection, the lung tissues of mice were taken for HE staining. The concentrations of TNF-α and IL-1β in peripheral blood serum were detected by ELISA. Results: There was no abnormality in the lungs of normal mice and low mice. The high dose of Salvia miltiorrhiza injection group mice showed diffuse hemorrhage and inflammatory cell infiltration in lung tissue. Compared with normal group and low dose of Salvia miltiorrhiza injection group, the concentration of TNF-α and IL-1β in high dose Salvia miltiorrhiza injection group was significantly Increased (P <0.05). Conclusion: Danshen injection can cause lung injury in mice and induce inflammatory reaction.
