目的:建立糖尿病肾病大鼠模型,研究桑枝提取物对其血糖、血脂、血黏度、尿微量白蛋白、血浆内皮素等指标及肾脏形态方面的影响。方法:采用单侧肾脏切除后腹腔注射链脲佐菌素(STZ)的方法复制糖尿病肾病大鼠模型,随机分为模型组、西药组、桑枝提取物低及高剂量组,每组10只,并设立正常组大鼠10只,灌胃给药14 d后测定血糖(FPG)、血脂(TC、TG、LDL-c、HDL-c)、血黏度(ηb、ηp、fib)、尿微量白蛋白(mAlb)、血浆内皮素(ET),并制作肾脏病理切片观察。结果:与单纯模型组比较,桑枝提取物治疗组FPG、TC、TG、LDL-c降低(P<0.05);HDL-c水平升高(P<0.05);ηb、ηp、fib下降(P<0.05);mAlb下降(P<0.05),其中FPG、TC、LDLc、HDL-c、ηb、ηp、mAlb高剂量组作用强于低剂量组(P<0.05);ET改善不明显(P>0.05);肾脏病理切片显示高剂量组大鼠肾脏损害轻于模型组。结论:桑枝提取物对DN模型大鼠的肾脏有一定的保护作用。
OBJECTIVE: To establish a rat model of diabetic nephropathy and study the effects of mulberry branch extract on blood sugar, blood lipid, blood viscosity, urine microalbumin, plasma endothelin and kidney morphology. Methods: The model of diabetic nephropathy was duplicated by intraperitoneal injection of streptozotocin (STZ) after unilateral nephrectomy. The model rats were randomly divided into model group, western medicine group and mulberry extract low and high dose group, with 10 rats in each group , And 10 rats in normal group were established. Blood glucose (FPG, TC, TG, LDL-c and HDL-c), blood viscosity (ηb, ηp, fib) Albumin (mAlb), plasma endothelin (ET), and making kidney pathological sections. Results: Compared with the model group, the levels of FPG, TC, TG and LDL-c in the mulberry branch extract group were significantly decreased (P <0.05), HDL-c levels were higher (P <0.05); mAlb decreased (P <0.05), and the effects of FPG, TC, LDLc, HDL-c, ηb, ηp and mAlb in high dose group were stronger than those in low dose group 0.05). Kidney pathological sections showed that the renal damage in high-dose group was lighter than that in model group. Conclusion: Mulberry extract has a protective effect on the kidneys of DN model rats.
