我们从基因库中筛选出乙型肝炎病毒(HBV)高度保守序列片段,通过分子克隆操作,用芯片点样仪点到特制的玻片上,制成乙型肝炎基因诊断芯片,检测了99例乙型肝炎肝硬化患者肝组织标本,同时用免疫组织化学法、原位分子杂交法检测乙型肝炎核心抗原(HBcAg)和HBV DNA。1.资料与方法:(1)芯片制备:玻片购自Sigma公司,HBV DNA靶基因和对照植物基因由博华公司提供,用通用引物进行PCR扩增,PCR产物用Cartesia7500点样仪进行点样制成芯片。(2)标本处理和DNA提取:采集本院病理科1995~2000年乙型肝炎后肝硬化肝活检
We screened out highly conserved sequence fragments of hepatitis B virus (HBV) from the gene pool. Through the molecular cloning operation, we spotted the sample onto a special glass slide to make a Hepatitis B gene diagnostic chip, and detected 99 cases Hepatic cirrhosis patients with liver tissue samples, at the same time by immunohistochemistry, in situ hybridization detection of hepatitis B core antigen (HBcAg) and HBV DNA. 1.
