本文报道含有脊髓灰质炎病毒Ⅰ型中和抗原位点及甲型肝炎病毒中和抗原位点重组痘苗病毒的构建,用重组痘苗病毒感染143TK-细胞后成功地表达了脊髓灰质炎病毒Ⅰ型和甲型肝炎病毒的抗原。免疫荧光显示,两种抗原的荧光颗粒均分布在胞浆内。Westernblot证实,重组痘苗病毒所表达的脊髓灰质炎病毒Ⅰ型抗原和甲型肝炎病毒抗原为一融合蛋白。用此重组痘苗病毒免疫动物,可诱导脊髓灰质炎病毒Ⅰ型和甲型肝炎病毒的中和抗体产生。
This article reports the construction of a recombinant vaccinia virus containing the type I neutralizing antigenic site of poliovirus and the neutralizing antigenic site of hepatitis A virus. The recombinant vaccinia virus was used to infect 143TK-cells and successfully expressed poliovirus type I and Hepatitis A virus antigen. Immunofluorescence showed that the fluorescent particles of both antigens were distributed in the cytoplasm. Westernblot confirmed that the recombinant vaccinia virus expressed poliovirus type I antigen and hepatitis A virus antigen as a fusion protein. Immunization of animals with this recombinant vaccinia virus can induce the production of neutralizing antibodies to poliovirus type I and hepatitis A virus.
