目的探讨农村老年人群硒水平与载脂蛋白E(APOE)基因多态性的关系。方法选择环境硒水平为适宜(邛崃市)和较低(剑阁县)的2地农村,用整群抽样方法,抽取自愿接受调查的65岁以上老人各500名。采集指甲样品进行硒含量测定;用特定滤纸采集指血检测APOE基因;另取50名自愿采血老人抽静脉血测硒。以SPSS软件和卡方分析比较老年人群硒水平与APOE基因多态性的差异。结果该2地老年人的血硒含量分别为(0.121±0.034)μg/g和(0.053±0.009)μg/g;指甲硒含量各为(0.457±0.203)μg/g及(0.201±0.038)μg/g,差异有统计学意义(P<0.01),并呈良好的正相关(r=0.496,P<0.05)。不同硒水平环境下长期居住老人的APOE基因多态性分布频率存在多水平和多层次的差别,机体硒水平与ε2等位基因的频率也有正相关。结论该2地农村老年人群体内硒水平不同,长期不同环境硒水平暴露,对APOE基因多态性的分布存在影响,机体内硒营养状况与APOE基因表达特别是ε2等位基因的表达存在关联。
Objective To investigate the relationship between selenium levels and apolipoprotein E (APOE) gene polymorphisms in rural elderly population. Methods A cluster sampling method was used to select 500 rural residents over the age of 65 who were volunteered to be surveyed in two rural areas with suitable environmental selenium levels (Wulong Municipality) and lower (Jiange County). Collect nails samples for determination of selenium content; with a specific filter paper refers to the finger detection APOE gene; another 50 voluntary blood collection of elderly venous blood selenium. SPSS software and chi-square analysis were used to compare the differences of selenium level and APOE gene polymorphism in the elderly. Results The blood selenium contents in the two geriatric adults were (0.121 ± 0.034) μg / g and (0.053 ± 0.009) μg / g, respectively. The finger selenium contents were 0.457 ± 0.203 μg / g and 0.201 ± 0.038 μg / / g, the difference was statistically significant (P <0.01), and showed a good positive correlation (r = 0.496, P <0.05). There was a multi-level and multi-level difference in the frequency of APOE gene polymorphism among long-term residents in different selenium levels. The selenium level was also positively correlated with the frequency of ε2 allele. Conclusion The selenium levels of the two rural elderly population are different, and long-term exposure to selenium in different environments has an impact on the distribution of APOE gene polymorphism. The relationship between selenium nutrition status and APOE gene expression, especially ε2 allele, is correlated with the distribution of APOE gene.
