目的观察糖肾宁对自发性2型糖尿病KK-Ay小鼠肾组织血小板衍生生长因子(platelet-derived growth factor,PDGF)、血小板衍生生长因子受体(platelet-derived growth factor receptor,PDGFR)蛋白表达的影响。方法采用自发性2型糖尿病KK-Ay小鼠建立糖尿病肾病模型,随机分为模型组、缬沙坦组和糖肾宁组各20只,设立C57BL/6J小鼠20只为空白组;药物干预12周后,测定24小时尿微量白蛋白排泄率(urinary albumin excretion,UAER),Masson染色观察各组肾脏病理,Western Blot检测PDGF、PDGFR蛋白表达水平。结果与空白组比较,模型组24小时UAER明显升高(P<0.05),肾组织PDGF、PDGFR蛋白表达明显升高;与模型组比较,缬沙坦组和糖肾宁组24小时UAER均有不同程度的降低(P<0.05),给药组间无显著性差异(P>0.05),肾组织PDGF、PDGFR蛋白表达有不同程度降低(P<0.05)。结论糖肾宁能够降低自发性2型糖尿病KK-Ay小鼠24小时UAER,抑制PDGF及PDGFR蛋白的过度表达,保护肾功能。
Objective To observe the effects of Tangshenning on protein expression of platelet-derived growth factor (PDGF) and platelet-derived growth factor receptor (PDGFR) in kidney of KK-Ay mice with spontaneous type 2 diabetes mellitus Impact. Methods Diabetic nephropathy model was established in KK-Ay mice with spontaneous type 2 diabetes mellitus, which were randomly divided into model group, valsartan group, and Tangshenning group, and 20 C57BL / 6J mice were established as blank group. After 12 weeks, urinary albumin excretion (UAER) was measured at 24 hours. Masson staining was used to observe renal pathology. Western Blot was used to detect PDGF and PDGFR protein expression. Results Compared with the blank group, UAER in model group was significantly increased at 24 hours (P <0.05), and the expression of PDGF and PDGFR in renal tissue was significantly increased. Compared with model group, 24-hour UAER in valsartan group and the control group (P <0.05). There was no significant difference between the two groups (P> 0.05). The expression of PDGF and PDGFR in renal tissue decreased to some extent (P <0.05). Conclusion Tangshenning can reduce 24-hour UAER, inhibit PDGF and PDGFR overexpression and protect renal function in spontaneous KK-Ay mice.
