高通量测序分析自身免疫性疾病T细胞受体β链CDR3组库的特征

自身免疫性疾病是机体对自身抗原发生免疫反应而导致自身组织损害所引起的疾病。导致其发病的基本因素可能是自身免疫耐受的破坏。T细胞在自身免疫病的适应性免疫应答过程中起着至关重要的作用。每一个成熟的 T 细胞表面表达一种特定的T细胞受体(TCR),互补决定区3(CDR3)最能直接反应TCR多样性。 T 细胞对众多抗原的识别依赖于CDR3区的多样性,故对TCR CDR3区序列的分析,可作为判别 T 细胞抗原识别特性的重要指标。研究[1]发现高通量测序技术检测 TCR CDR3组库特征变化已被广泛应用,包括恶性肿瘤、自身免疫性疾病和感染性疾病等。本文就自身免疫性疾病的TCRβ链CDR3组库特征变化的研究进展进行综述。
