目的:探讨逍遥散对雌性兔膝关节软骨细胞超微结构的影响。方法:采用改良Huith法对30只雌性新西兰兔右膝关节进行膝骨关节炎造模,造模1周后将30只实验兔随机分为逍遥丸组、玻璃酸钠组及对照组,每组10只。逍遥丸组每天每只给予8丸逍遥丸(相当于原药材3 g),配成30 mL液体灌胃,连用5周;玻璃酸钠组予以玻璃酸钠注射液0.15 mL.kg-1右膝关节内注射,每周1次,连用5周;对照组每天予以30 mL生理盐水灌胃,连用5周。造模后12周末处死所有实验兔,取右膝内侧胫骨平台软骨组织切片后透射电镜观察。结果:逍遥丸组细胞核正常,核膜完整,核质均匀,线粒体嵴部分模糊,出现空泡化现象,内质网、高尔基体正常,纤维组织稍紊乱,有凋亡小体存在;玻璃酸钠组细胞核正常,核膜完整,核质均匀,线粒体空泡化较轻,内质网、高尔基体正常,纤维组织稍紊乱,有凋亡小体存在;对照组细胞核可见固缩现象,线粒体异常,表现为线粒体嵴模糊、消失,空泡化现象较明显,内质网、高尔基体基本正常,纤维组织紊乱现象较明显,多个标本见凋亡小体。结论:逍遥散对雌性兔膝关节软骨细胞具有保护作用。
Objective: To investigate the effect of Xiaoyaosan on the ultrastructure of rabbit articular chondrocytes. Methods: Thirty knee osteoarthritis of New Zealand rabbits were made by modified Huith method. One week later, 30 rabbits were randomly divided into Xiaoyao Pill group, sodium hyaluronate group and control group. Each group 10 only Xiaoyao bolus group given 8 pills Xiaoyao Pill every day (equivalent to the original medicinal 3 g), dubbed 30 mL liquid gavage for 5 weeks; sodium hyaluronate sodium injection 0.15 mL.kg-1 right knee Intra-articular injection, once a week, once every 5 weeks; the control group was given 30 mL saline daily gavage for 5 weeks. All experimental rabbits were sacrificed at the end of 12 weeks after modeling, and the cartilage tissue sections of the medial tibial plateau in the right knee were observed by transmission electron microscopy. Results: Xiaoyao Wan had normal nucleus, complete nuclear membrane, uniform nucleus, partially blurred mitochondrial cristae, vacuolization, normal endoplasmic reticulum and Golgi apparatus, slightly disorganized fibrous tissue and presence of apoptotic bodies. Sodium hyaluronate The normal nucleus of nucleus, nucleus were homogeneous, mitochondria vacuolization was mild, endoplasmic reticulum and Golgi apparatus were normal, fibrous tissue was slightly disordered and apoptotic bodies existed. In the control group, nuclear condensation, mitochondria abnormality, Manifested as mitochondrial cristae blurred, disappear, vacuolization phenomenon is more obvious, endoplasmic reticulum, Golgi apparatus was normal, fibrous tissue disorders more obvious, many specimens see the apoptotic body. Conclusion: Xiaoyaosan has a protective effect on rabbit knee chondrocytes.
