目的:建立荷叶提取物中黄酮类成分的高效液相测定方法。方法:药材经提取水解之后用HPLC通过测定其槲皮素的含量来确定荷叶提取物中总黄酮的含量,采用Dia-monsil C18色谱柱(4.6mm×250mm,5.0μm),乙腈-0.4%磷酸(35:65)为流动相;流速1.0mL.min-1;检测波长360nm;柱温25℃。结果:槲皮素在30~180μg范围内呈良好的线性关系(r=0.9996),平均回收率为96.19%,RSD为1.75%(n=5)。结论:此测定方法简便、准确、重复性好,可以作为荷叶药材中总黄酮的含量测定方法,以控制荷叶药材的质量。
Objective: To establish a method for the determination of flavonoids in lotus leaf extract by high performance liquid chromatography. Methods: The content of total flavonoids in lotus leaf extract was determined by HPLC after being extracted and hydrolyzed by HPLC. The content of total flavonoids in lotus leaf extract was determined by using Dia-monsil C18 column (4.6 mm × 250 mm, 5.0 μm), acetonitrile-0.4% Phosphoric acid (35:65) as mobile phase; flow rate 1.0mL.min-1; detection wavelength 360nm; column temperature 25 ℃. Results: Quercetin had a good linearity (r = 0.9996) in the range of 30-180 μg with an average recovery of 96.19% and a RSD of 1.75% (n = 5). Conclusion: The method is simple, accurate and reproducible. It can be used as a method for the determination of total flavonoids in lotus leaf in order to control the quality of the lotus leaf.
