本实验以石竹带芽茎段为起始材料,通过对 MS 培养基加入不同激素种类和浓度梯度的处理,观察外植体在愈伤组织的产生、苗的分化、根的生长和侧枝萌发等方面产生差异,从而筛选出石竹组织培养过程中不同阶段适宜的培养基。结果表明:1)石竹诱导愈伤组织适宜培养基为 KT0.05mg·L~(-1)+NAAI.0mg·L~(-1);2)诱导分化苗适宜培养基为 KT0.1mg·L~(-1)+NAA1.0mg·L~(-1);3)诱导生根适宜培养基为 NAA2.0mg·L~(-1);4)诱导大量侧枝萌发适宜培养基为 BA4.0mg·L~(-1)+NAA2.0mg·L~(-1)。5)外源激素对石竹试管花形成不是必须的因子。6)对于试管苗的移植选择适宜基质为泥炭+珍珠岩。
