目的:观察人参强心滴丸对慢性心衰大鼠血浆AngⅡ含量与心肌组织AngⅡ受体AT1 mRNA、PKC表达的影响,揭示人参强心滴丸治疗慢性心衰的作用机制。方法:采用腹主动脉缩窄法复制慢性心衰大鼠模型,随机分为假手术组、模型组、阳性药对照组和人参强心滴丸高、低剂量组。检测大鼠血浆AngⅡ的含量和心肌组织PKC以及AT1 mRNA的表达变化。结果:模型组大鼠血浆AngⅡ含量明显增加,心肌组织中AT1 mRNA和PKC的表达明显增多;给药后,各治疗组大鼠AngⅡ的含量明显减少,AT1 mRNA和PKC的表达明显降低。结论:人参强心滴丸能降低AngⅡ的含量,抑制AT1 mRNA和PKC的表达,可能是其治疗慢性心衰的分子机制之一。
Objective: To observe the effect of ginseng Qixin Diwan on Angiotensin Ⅱ content and Angiotensin Ⅱ Receptor (AT1) mRNA and PKC expression in chronic heart failure rats and to reveal the mechanism of ginseng Qixin Dripping Pills in treating chronic heart failure. Methods: A rat model of chronic heart failure was established by abdominal aorta stenosis. The rats were randomly divided into sham operation group, model group, positive control group and ginseng QXD pills in high and low dose groups. The contents of AngⅡ and the expressions of PKC and AT1 mRNA in myocardium were detected. Results: The content of AngⅡ in the model group was significantly increased, and the expression of AT1 mRNA and PKC in myocardium was significantly increased. After treatment, the content of AngⅡ and the expression of AT1 mRNA and PKC in the treatment group were significantly decreased. Conclusion: Ginseng Qixin Drop Pill can reduce the content of Ang Ⅱ and inhibit the expression of AT1 mRNA and PKC, which may be one of the molecular mechanisms of its treatment of chronic heart failure.
