目的探讨孟鲁司特对脓毒症大鼠肺组织半胱氨酰白三烯受体1(CysLT1R)表达的影响。方法将48只SD大鼠随机均分成模型组及孟鲁司特治疗组。用盲肠结扎穿孔及导管留置法制备脓毒症急性肺损伤大鼠模型。治疗组术前30min用孟鲁司特钠咀嚼片按10mg·kg-1·d-1剂量稀释后给大鼠灌胃,持续至实验结束。两组分别在术后6、12、24、48h分别处死6只大鼠,取肺组织;用免疫组化法检测肺组织CysLT1R的表达,RT-PCR法检测肺组织CysLT1RmRNA表达,并观察肺组织的病理变化。结果各时间点治疗组大鼠肺组织CysLT1RmRNA和CysLT1R表达均低于模型组(P<0.05)。治疗组肺组织病理损伤较模型组减轻(P<0.05)。结论孟鲁司特能下调脓毒症大鼠肺组织CysLT1R的表达,对脓毒症大鼠肺损伤起到保护作用。
Objective To investigate the effect of montelukast on the expression of cysteinyl leukotriene receptor 1 (CysLT1R) in lung tissue of septic rats. Methods 48 SD rats were randomly divided into model group and montelukast treatment group. Preparation of septic acute lung injury rat model by cecal ligation and perforation and catheter indwelling. The rats in the treatment group were intragastrically administrated with montelukast sodium chewable tablets at a dosage of 10 mg · kg-1 · d-1 for 30 min before the experiment was finished. Six rats were sacrificed at 6, 12, 24, and 48 h after operation, respectively. The lungs were harvested. The expression of CysLT1R in lung tissue was detected by immunohistochemistry. The expression of CysLT1R mRNA in lung tissues was detected by RT-PCR. Pathological changes. Results The expression of CysLT1R mRNA and CysLT1R in the lung tissue of the treated group were lower than those of the model group at each time point (P <0.05). Compared with the model group, the pathological changes of lung tissue in treatment group were alleviated (P <0.05). Conclusion Montelukast can downregulate the expression of CysLT1R in the lung tissue of septic rats and protect lung injury in septic rats.
