本文报道将抗日本血吸虫循环阴极抗原单克隆抗体(McAbⅢD_(10)),分别用于检测宿主血清中循环抗原和抗CCA特异性抗体。以McAb ⅢD_(10)建立的竞争性ELISA对诊断血吸虫病有高度的敏感性和特异性,极少交叉反应。用单盲法检测了三批样本,符合率均较满意。用此法检测1915份样本,其中急性血吸虫病113例,100％阳性;慢性血吸虫病765例,阳性率96.3％;晚期血吸虫病25例,阳性率72％;血吸虫病治愈者66例,阴转率70％,正常人750例,均阴性;包虫病27例,无交叉反应;并殖吸虫病43例及华支睾吸虫病126例,分别有1和2例阳性。用此单抗建立的Dot-ELISA,具有疗效考核价值,血吸虫病治愈者48例,阴转率97.9％。动物实验中,治后8wk,16只治愈家兔的阴转率为76％,其余4只阳性者,阳性滴度和阳性强度均降低。C-ELISA和Dot-ELISA两者的符合率较好,若一份血样同时用此两种方法检测可以互补。
In this paper, monoclonal antibody against the antigen of Schistosoma japonicum circulating antigen (McAbⅢD_ (10)) was used to detect circulating antigens and anti-CCA specific antibodies in the serum of the host. The competitive ELISA established with McAb Ⅲ D_ (10) is highly sensitive and specific for the diagnosis of schistosomiasis with minimal cross-reaction. Three batches of samples were tested by single blind method, and the coincidence rate was satisfactory. 1915 samples were detected by this method, of which 113 cases were acute schistosomiasis, 100% positive; 765 cases were chronic schistosomiasis, the positive rate was 96.3%; 25 cases were advanced schistosomiasis, the positive rate was 72%; 66 cases were cured by schistosomiasis, 70% of normal, 750 cases of normal, were negative; echinococcosis in 27 cases, no cross-reaction; 43 cases of paragonimiasis and Clonorchiasis 126 cases, respectively, 1 and 2 were positive. The Dot-ELISA established with this monoclonal antibody has the value of curative effect evaluation, 48 cases of schistosomiasis cured, the negative conversion rate of 97.9%. Animal experiments, 8wk after treatment, 16 rabbits were cured of the negative conversion rate was 76%, the remaining 4 were positive, the positive titer and positive intensity were reduced. The coincidence rate between C-ELISA and Dot-ELISA is good, and if a blood sample is detected by both methods, it can be complementary.
