目的:通过相溶解度实验研究葛根素与羟丙基-β-环糊精(hydroxypropyl-β-cyclodextrin,HPCD)的包合作用。采用热力学方法阐明葛根素的包合机制。方法:用冷冻干燥法制备葛根素/HPCD包合物,采用DSC、扫描电镜和红外光谱仪对固体包合物进行了表征,并测定了葛根素在HPCD水溶液中的包合常数。结果:葛根素与HPCD可形成1∶1(物质的量比)的包合物,通过计算包合过程的熵变、焓变及自由能变化,得出升高温度有利于包合反应的进行。结论:葛根素-HPCD的包合过程是一个吸热的自发过程。
OBJECTIVE: To study the inclusion effect of puerarin and hydroxypropyl-β-cyclodextrin (HPCD) by phase solubility experiments. The inclusion mechanism of puerarin was elucidated by thermodynamic method. Methods: The puerarin / HPCD inclusion complex was prepared by freeze-drying method. The inclusion complex was characterized by DSC, SEM and FTIR. The inclusion complex of puerarin in HPCD solution was determined. Results: The inclusion complex of puerarin with HPCD could be formed by 1: 1 (mass ratio). By calculating the entropy change, enthalpy change and free energy change during the inclusion process, it was concluded that the increase of temperature was in favor of the inclusion reaction . Conclusion: The inclusion process of puerarin-HPCD is an endothermic spontaneous process.
